Gene synthesis method

The methods of gene synthesis cannot be substituted for each other, and each method can be applied in different experiments. 

Here is an overview of several common gene synthesis methods:

Solid phase synthesis

Traditional oligonucleotide synthesis uses a microliter volume of solution for synthesis on a small column. 

Oligonucleotide synthesis involves adding nucleotide monomers one by one at the end of the synthetic chain. 

The synthesis of each oligonucleotide contains four steps: deprotection, coupling, blocking and oxidation. 

Sequence integrity and synthetic productivity hinder product lengths longer than 200bp, so this method generally limits the length of DNA sequences. 

The main advantage of this method is its high accuracy, which also makes people accept the high cost and low output of this method.

Chip DNA synthesis

As the name suggests, chip DNA synthesis is a technology that uses a series of electrochemical techniques to synthesize DNA using a microarray chip. 

Different kinds of nucleotides can be synthesized at different specific positions on the chip, which are called oligonucleotide pools. Following this segmented synthesis method, gene fragments are amplified in the oligo-nucleic acid pool, and then aggregated and assembled into the finished product. 

The chip DNA synthesis method is cheaper than solid-phase synthesis and can produce a larger number of target genes, but the accuracy of this method is comparable.

PCR synthesis

The gene fragments generated by the PCR method can be completed by various cell systems. For example, using the yeast system, different genes can be added to the yeast chromosomes by using different nick labeling enzymes and tags. 

Since it has the characteristics of being able to insert genes into the target genome, there is no length limit to the extent that the chromosome can accommodate. 

This method has a good effect when synthesizing long gene fragments. With the help of the cell system, the accuracy of the gene sequence can be guaranteed.
